塔里木兔是新疆重要的野生保护动物之一
而性别鉴定是开展野兔种群现状、群体遗传学和生态学研究的基础和前提工作.文章以新疆喀什地区85例未知性别的塔里木兔样本为研究对象
样本类型包括血液、肌肉、皮张等
通过改良的酚-氯仿法提取高质量的野兔基因组DNA
利用兔类SRY(Sex Determining Region Y)和APP(Amyloid Precursor Protein)基因的两对特异性引物
采用多重PCR的方法对其性别进行鉴定
成功鉴定出了43例雌性
42例雄性塔里木兔
雌雄比例为1.02∶1
说明了该地区塔里木兔性别比例平衡
野兔群体具有较强的繁殖潜力.本研究中的多重PCR方法可以对塔里木兔的性别进行快速、准确地鉴定
获得的实验结果可为今后塔里木兔的生物学研究和保护利用等提供基础数据.
Yarkand Hare is one of the important protected wildlife in Xinjiang. Sex determination is the basis and prerequisite for the study of population status
population genetics and ecology of hares. In this study
85 samples of sex-unknown Yarkand Hare from Kashgar region of Xinjiang
including blood
muscles and skins
etc.
were involved. High quality genomic DNA of hares was extracted with modified phenol-chloroform method. The specific primers of SRY and APP gene in Leporidae were used to determine the sex of Yarkand Hare by multiplex PCR. 42 males and 43 females were identified successfully. The ratio of male to female is 1∶1.02. It shows that the sex ratio of Yarkand Hare is balanced in this area
and the population has strong reproductive potential. The multiplex PCR method in this study can quickly and accurately identify the sex of Yarkand Hare. The experimental results would provide basic data for the future biological research
protection and utilization of Yarkand Hare.
