比较新疆一枝蒿未除多糖醇提物(Ethanol extract of wild Artemisia rupestris L.

DEEWAR)的免疫调节活性.利用体外树突状细胞(Dendritic cells
DCs)为平台
FACS (Fluorescence Activating Cell Sorter)技术检测未除多糖EEWAR和除多糖EEWAR对DCs成熟的影响.超声法提取EEWAR
检测其多糖和黄酮含量.C57BL/6小鼠骨髓细胞进行体外培养
第7 d后收获DCs
不同剂量未除多糖及除多糖的EEWAR分别刺激DCs细胞12 h后
LPS为阳性对照
检测DCs表面分子(CD11c
CD40
CD80
CD86和MHCII)表达情况.未除多糖和除多糖的EEWAR多糖含量分别为30.1%
5.3%
黄酮含量分别为5.6%
16.4%.FACS实验结果显示
未除多糖及除多糖的EEWAR均能够剂量依赖性促进CD80
CD86和MHCII分子的表达水平
且除多糖后促进DCs成熟的能力增强了约一倍;未除多糖及除多糖的EEWAR对CD40分子的促进作用不明显.结果表明
EEWAR除多糖后免疫调节活性明显增强.
To compare the immunoregulatory activities of ethanol extract of wild Artemisia rupestris L.
(EEWAR)and ethanol extract of wild Artemisia rupestris L.(DEEWAR) removing polysaccharides. Using Dendritic cells(Dendritic cells
DCs) as a platform in vitro
The effects of EEWAR with polysaccharide or polysaccharide removal on DCs maturtion were detected by Fluorescence Activating Cell Sorter(FACS). EEWAR was extracted by ultrasonic method and its content of sugar and flavonoid was detected. Marrow-derived cells from C57BL/6 mice were cultured in vitro
and DCs were harvested on day 7 after culture. DCs were stimulated for 12 h by EEWAR with different doses
respectively. Expression of DCs surface molecules(CD11c
CD40
CD80
CD86 and MHCII)were detected. The content of polysaccharides and flavonoid of EEWAR was 30.1%
5.3%
5.6% and 16.4%
respectively. FACS results showed that EEWAR with polysaccharide or polysaccharide removal could increase the expression levels of CD80
CD86 and MHCII molecules in a dose-dependent manner
and the ability to promote DCs maturation after removing polysaccharides was doubled. The effect of EEWAR on CD40 was not obvious.The results showed that the immunomodulatory activity of EEWAR was significantly enhanced after the removal of polysaccharides.
