The activities of cellulase by Nitrogen ion implantation of different dosage on the bacillus. sp HY2 are studied in order to choose the high-activity mutant. The results show bacillus. sp HY2-3 is the high-activity mutant. The chy2 encoding bacillus
sp HY2 and the chy2-3 encoding bacillus and sp HY2-3 are amplified by PCR
the two amplified fragment are cloned and sequenced. After being digested by PstI
chy2-3 and chy2 fragments are linked with vector PGEM-T and transformed into competent TG-1. Positive clones are screened
plasmid PGEM-T/chy2 and plasmid. PGEM-T/ chy2-3 are detected by PCR and restriction digestion. The results show that two coding regions all comprise 1 500 nucleotides and that there are some mutation in the sequences of chy2 and chy2-3
which have effect on encoding the ploypeptide of amono acids and the activities of cellulase.
