以甜瓜品种红蜜脆、皇后的无菌子叶块为外植体
用农杆菌介导法进行遗传转化处理.针对遗传转化处理后
在外植体上长出的甜瓜不定芽发生黄化的现象
从品种差异、遗传转化条件、矿物营养含量、光照强度、温度等多方面作了初步探讨.结果表明:与未遗传转化处理的子叶块上长出的不定芽相比
转化处理后的子叶块上长出的不定芽黄化现象普遍.将后者分别转接入多添加了Fe或Mg或B或K盐的改良芽生长培养基上培养
一段时间后
形态观察表明
在多添加了Fe盐的培养基上不定芽转绿明显
其次
在多添加了Mg盐的培养基上不定芽也有转绿.显微、超微结构上显示
在多添加了Fe盐的培养基上不定芽叶栅栏细胞内含较多的叶绿体
其叶绿体内有丰富的光合片层结构.光照强度对不定芽转绿作用不明显
温度保持恒定(270C+20C)或波动(150C~350C)对甜瓜不定芽转绿影响不大
两个品种在这方面的差异很小.
This paper reported that the explants of bacteria-free cotyledons of Xinjiang muskmelon was trans-geneted treatment through Agrobacterium tumefaciens. Be directed against the yellowing leaves of adventitious buds after trangenic treatment
we engaged explorations from strain variance and conditions of genetic transformation and contents of mineral nutritious and intensity of illumination as well as temperation. The results indicated that the adventitious buds
from the explants of transgenic treatment
are yellowing universally by contrast. We took them respectively on the improve mediums of bud-growth which added Fe or Mg or B or K salt. After a period of time
the results indicated from morphological observation that the adventitious buds grew on the medium added Fe salt became green and green evidently
and so did the bud on the medium added Mg salt. The results indicated from micro and super-micro structures that the palisade cells of adventitious buds from medium added Fe salt contain many more chloroplasts
and also there are a lot of photosynthetic lamellaes are rich in the chloroplast.
