考察共载姜黄素和甘草酸的介孔硅敷料在蓝光照射下用于烫伤创面修复的潜力及初步作用机制.基于吸水特性检测敷料吸收创面渗出液潜力,根据创面愈合率、体重变化和病理切片检测评价敷料促进创面愈合能力,通过检测创面处血管内皮生长因子VEGF(Vascular Endothelial Growth Factor)的变化评价敷料促进血管生成功效、检测肝脏中的炎症因子白介素IL-1β(Interleukin-1β)评价对肝脏修护作用,利用在体抗菌活性检测敷料抗菌效果并通过对活性氧(ROS)的检测分析其抗菌机制.结果表明:载药后的微米介孔硅不影响本身的吸水特性.蓝光照射下对创面愈合率、病理特征、创面处抑制金黄色葡萄球菌能力、创面处VEGF和肝脏中IL-1β均同模型组有显著差异(P <0.05),其中抑菌能力可能来源于活性氧的产生.蓝光照射能缓解载药敷料对烫伤小鼠体重的降低(P <0.05).蓝光能协同共载姜黄素和甘草酸的介孔硅敷料用于小鼠烫伤创面的恢复.
To investigate the possibility and preliminary mechanism of μm-sized mesoporous silica(μm S) codelivering with curcumin(Cur) and glycyrrhizic acid(GA) as a wound-dressing(Cur+GA@μm S) for scald wound repair under blue light irradiation. The ability of the wound-dressing to absorb wound exudate was detected by water absorption characteristics. The ability of the wound-dressing to promote wound healing was evaluated by wound healing rate
body weight change and pathological section detection. The effect of the wound-dressing on angiogenesis was evaluated by detecting the change of Vascular Endothelial Growth Factor(VEGF) at the wound.The repair effect on the liver was evaluated by detecting the inflammatory factor Interleukin-1β(IL-1β) in the liver.The antibacterial effect of the wound-dressing was detected by in vivo antibacterial activity and was explaned by reactive oxygen species(ROS) production. VEGF at the wound and IL-1β in the liver caused by Cur+GA@μm S were significantly different from those in the model group(P <0.05). ROS production may be the reason of the inhibiton to staphylococcus aureus of Cur+GA@μm S. Cur+GA@μm S combined with blue light irradiation could alleviate the weight loss of scalded mice caused by the former(P <0.05). Cur+GA@μm S in photodynamic therapy with blue light can be used for the recovery of scald wounds in mice.
